Does this take place on LV-426, the planet from Alien and Aliens?
No it does not. it takes place on LV-223 which is another moon planet but has better conditions for sustaining life than LV-426 did before it was terra-formed. It could be said that LV-426 is in the same star system as LV-223 "Zeta II Reticuli" although the solar systems seen in Alien and Prometheus may still lie several or hundreds of light years apart. In "Alien", Lambert mentions they are just short of "Zeta II Reticuli" when they are trying to figure out where they are, however, the Nostromo also heads toward a moon surrounding a ringed planet during the landing sequence in the film similar to the one in Prometheus. The 3D imaging shows the ringed planet with "Zeta II Reticuli" on the screen as well. It should be noted that Zeta Reticuli is a star system, not a planetary one, therefore, both locations could be different but near one another, as Shaw's presentation in this film indicates that this ringed planet has one sole moon.
How is this related to Ridley Scott's 'Alien' and the rest of the series?
The movie is set in the same fictional universe of Ridley Scott's Alien and has shared events with it, and directly precedes the rest of the original series, but it's definitively not related to Aliens vs Predator. There had been talks of a fifth Alien movie for years, with alternating contributions from James Cameron and Ridley Scott. In fact, Scott had committed to an Alien sequel or prequel several years ago but aborted the project when the studio opted to make the AvP movies first, and both Cameron and Scott, separately, were not supporters of the Alien/Predator crossover series. After the disappointing box office from Aliens vs Predator: Requiem, the studio shelved the AvP series again to go back to the original plan. The prequel film was supposed to cover the origins of the Weyland-Yutani Company.
When the project was initially greenlit, it was seen as two separate movies to be chronologically before the first Alien movie, written by Jon Spaihts, being directed by Carl Erik Rinsc, and Scott staying only as producer. However, Fox wanted Scott in the director's chair so much that he eventually accepted under the condition that the script was heavily retooled to reflect the concepts that he wanted to explore. Damon Lindelof was the pen to make it happen, and the result has been described in certain moments as a both a prequel and an original story. For instance, in an interview, Michael Fassbender confirmed that this is actually an Alien movie, taking place in the Alien universe. In July 2011, however, Scott stated in another interview that "by the end of the third act you start to realize there's a DNA of the very first Alien, but none of the subsequent [films]".
In short, the story takes place within the same universe albeit not directly connected to the events of the original. There has been some misdirection on part of the film makers to help heighten interest in the film by showing glimpses and links in common with Alien, including imagery inspired by, if not designed and built by, H.R. Giger (Source). Prometheus depicts that the so-called "Space Jockey" in Alien is a member of an ancient race nicknamed "Engineers" by the Prometheus Crew, and that they were aware of the existence of the race called in other films as "xenomorph", but never clearly explains the exact relationship between both races. It's only hinted that the "xenomorph" is used as a weapon or some other device to control less evolved races.
Was the ship from this film the one found in Alien?
No, the Derelict ship found in Alien was found on the planet LV-426 (henceforth called Derelict-426). The ship in Prometheus crashes on planet LV-223 (Derelict-223) after colliding with the Prometheus in the year 2093. Captain Janek theorizes that LV-223 is a military base, and this is reinforced when David tells Elizabeth that there is more than one ship and they are seen leaving the planet in one. In Prometheus, we learn that the last Engineer survived some kind of outbreak aboard Derelict-223 and never took off as it is in its docking station. We also see that the cargo of Derelict-223 is similar in function to Derelict-426, however different in appearance and maybe purpose as well. When the crew find the decapitated body of one of the Space Jockeys, they deduce it had been dead for at least 2000 years. In Alien, Dallas states that the Space Jockey (as Engineers were called before this movie) "looks like it's been dead a long time. Fossilized. Looks like it's growing out of the chair." Prometheus takes place in late December 2093 and Alien takes place in 2122, so it's safe to assume that the Space Jockey (from Derelict-426) didn't fossilize in just 28 years, but instead fled LV-223 during the initial outbreak which killed off all the Engineers in the planet. Unbeknownst to it, it had been impregnated by its cargo and died during the birthing process, and Derelict-426 crashed on LV-426 over 2000 years ago and remained dormant until found by the crew of the Nostromo.
Where in Prometheus did they say that this planet was LV-223?
When they show the holographic pictogram of the ancient "invitations" on Prometheus they show it's a planetary system that contains LV-223.
